You may have heard of the so-called “Wolverine” peptide stack doing the rounds in fitness and wellness circles. But what exactly is it? And is it safe? We examine the evidence.
There is much debate over the safety and effectiveness of individual peptides. Only FDA-approved peptide medications (such as insulin and GLP-1s) have undergone clinical review for safety and effectiveness.
Peptides newly approved for compounding (such as BPC-157, KPV, and TB-500) have only been evaluated for specific use cases. They have not been approved by the FDA or extensively tested for safety or effectiveness, especially with ongoing use. Doctors and scientists say there is a lack of sufficient scientific evidence for the safety of non-FDA-approved injectable peptides marketed for wellness.
We’re keeping track of the developing regulatory and medical evidence around peptides, but it’s best to speak with your doctor or healthcare professional before introducing any peptide product.
Peptides are simply shorter forms of proteins and are made up of amino acids, just like proteins are. Your body produces some peptides naturally, and peptide drugs are often made to mimic these naturally occurring substances.
You might be familiar with the peptide drugs GLP-1s (glucagon-like peptide-1s). These drugs are approved by the FDA for specific medical conditions, and you’ll need a prescription to get them.
Scientists are also investigating peptides for potential uses in skin and muscle repair. You might come across peptides being sold for topical (on the skin) use as well as injectables for muscle regeneration.
Since the research is still in the early stages, these uses are not currently approved by the FDA and may carry risks. However, use on the skin is likely much safer than injecting these substances.
One example of an injectable muscle peptide is the so-called “Wolverine” peptide stack.
Peptide stacking is simply using more than one peptide at the same time, in the same formula. The so-called “Wolverine” peptide stack is a combination of two synthetic peptides:
- BPC-157 (Body Protection Compound 157)
- TB-500 (Thymosin Beta 500, which is a small part of Thymosin Beta-4)
This stack has been dubbed the “Wolverine” stack for its supposed regenerative effects.
Though fitness influencers may be touting the proposed healing benefits of this product, it is not FDA approved, as research never reached the clinical trial stage. Therefore, it has not been tested in humans, and neither effectiveness nor safety has been verified.
Reports of use in humans are purely anecdotal. And some side effects have been reported, including injection site irritation, stomach upset, lethargy, and headaches.
Though initial studies on the peptides in the Wolverine peptide stack appear promising, research remains in the early stages. And there’s usually a long way to go before a drug gets approved for use in humans.
Like any other drug, the peptides in the Wolverine stack should demonstrate safety and effectiveness before approval, and scientists typically also determine appropriate doses through an established, structured process.
Let’s take a look at what the research so far shows:
BPC-157
A 2025 review of preclinical (non-human) studies suggests a potential use of BPC-157 in improving musculoskeletal symptoms. However, the review notes that none of the studies showed clinical safety evidence — that means that all the studies were early stage and did not include human trials.
Another 2021 early stage animal study showed a potential for BPC-157 to reduce pain and increase muscle and nerve healing in rodents. However, this effect was only seen in one stage of the study and not the other.
Medical consensus is that before clinical trials in humans have been carried out, BPC-157 should not be taken by anyone for any purpose.
TB-500
Animal and in vitro (test tube) studies suggest that TB-500 may improve wound healing through its metabolite, Ac-LKKTE.
Again, as with BPC-157, these studies were not carried out in humans but in test tubes and animal models.
Findings in these types of studies may translate into humans, but high quality trials would be needed to determine if this is the case.

Proponents typically inject the formula, either subcutaneously or intramuscularly. However, this is not recommended by healthcare professionals and could lead to side effects, including irritation at the injection site, stomach issues, and other unknown effects.
Injecting is also much riskier than other administration routes, such as topical (applied to the skin) use, since it bypasses many protective immune barriers, such as those in the skin, gut, and lungs. Contamination of the injection needle is also a real risk when the conditions in which an unregulated drug is manufactured cannot be verified. Infection or the introduction of other harmful substances can also happen.
The risk of severe safety effects is also unknown. We don’t know whether these peptides affect the immune system or increase the risk of cancer, for example, because no human safety trials have been conducted. Since some peptides work by promoting the growth of new blood vessels to promote healing, they could theoretically promote the growth of new blood vessels that deliver nutrients to a tumor if a person already has precancerous cells.

Peptides in the Wolverine stack are currently not regulated in the United States. They are also not approved in any country and remain banned by the World Anti-Doping Agency.
When something sounds too good to be true, it likely is. And a drug referencing a Marvel character’s regenerative healing powers may just be exactly that.
Though many influencers, so-called “wellness experts”, and “biohackers” tout the benefits of the Wolverine peptide stack, research on using peptides in this way is very much preliminary and carries substantial risk of the unknown should you choose to try it.
That said, there are plenty of other ways to boost your fitness regimen.
